Certain widgets can only be added to certain zones.
"Why," you ask? Because we want profile pages to have freedom of customization, but also to have some consistency. This way, when anyone visits a deviant, they know they can always find the art in the top left, and personal info in the top right.
Don't forget, restraints can bring out the creativity in you!

There are 2 things "Design Guideline" can mean. Either a set of guidelines other people have to adhere to in order to use the logo appropriately/with your permission or explanations of the design (how it works, what it does well, how you came to that design, etc.)
"Also will you be able to produce a design guideline if you are the winner?"
That sounds to me like they're asking you to do the latter of the two and provide information about your design. If you're still confused, you may be able to get some hint of what they want by asking them to clarify.
